Sanctuary volgt de avonturen van de mooie en mysterieuze dr. Helen Magnus (Amanda Tapping) en haar team. Magnus hee één missie: de vreemde en angstaanjagende wezens genaamd ‘abnormals’ beschermen in haar heiligdom. Terwijl de rest van de wereld het bestaan van de wezens weigert te geloven, spoort Magnus de abnormals op.
In het tweede seizoen zet Magnus de zoektocht naar haar vermiste dochter Ashley voort. Hierbij komt ze erachter dat niet alleen haar dochter in gevaar is, maar dat het hele heiligdom wordt bedreigd.
